Como o NGS introduz um n\u00famero relevante de falsos positivos e falsos negativos, a CENTOGENE determinou como otimizar o uso do sequenciamento de Sanger ortogonal de variantes em paralelo ao analisar casos lim\u00edtrofes. Na edi\u00e7\u00e3o de janeiro de 2019 da revista Genetics in Medicine, a CENTOGENE descreveu, no artigo intitulado &quot;Desenvolvimento de um algoritmo baseado em evid\u00eancias que otimiza a sensibilidade e a especificidade no diagn\u00f3stico baseado em sequenciamento de exoma (ES) de uma popula\u00e7\u00e3o de pacientes clinicamente heterog\u00eanea&quot;, uma estrutura na qual as variantes identificadas pelo NGS podem ser avaliadas e monitoradas de forma consistente, reduzindo significativamente a necessidade dessa valida\u00e7\u00e3o adicional.<\/p>\n<p>A CENTOGENE desenvolveu uma \u00e1rvore de decis\u00e3o que considera tr\u00eas caracter\u00edsticas espec\u00edficas das variantes: pontua\u00e7\u00e3o de qualidade, n\u00famero de leituras e frequ\u00eancia. Ao utilizar esse preditor, \u00e9 poss\u00edvel classificar 91,71% das variantes com especificidade de 100% e sensibilidade de 99,8%, restando apenas 8,31% das variantes para confirma\u00e7\u00e3o por sequenciamento Sanger adicional. Esse resultado evita a subnotifica\u00e7\u00e3o e a supernotifica\u00e7\u00e3o, possibilitando diagn\u00f3sticos mais precisos para pacientes com doen\u00e7as raras que, de outra forma, n\u00e3o seriam detectadas. As implica\u00e7\u00f5es desse aumento na especificidade e na sensibilidade tamb\u00e9m se traduzem em capacidades de monitoramento mais consistentes.<\/p>\n<p>\u201cA nova estrutura que desenvolvemos para otimizar a sensibilidade e a especificidade em diagn\u00f3sticos baseados em sequenciamento de exoma completo (WES) refor\u00e7a o foco estrat\u00e9gico da CENTOGENE: transformar dados cl\u00ednicos e gen\u00e9ticos para impulsionar o diagn\u00f3stico m\u00e9dico r\u00e1pido de doen\u00e7as heredit\u00e1rias\u201d, disse o Professor Peter Bauer, MD, Diretor Cient\u00edfico da CENTOGENE e principal autor do estudo. \u201cEssa estrutura pode ser otimizada em toda a ind\u00fastria, academia e cl\u00ednica sem comprometer a precis\u00e3o do diagn\u00f3stico gen\u00e9tico.\u201d<\/p>\n<p>O estudo foi realizado em uma ampla popula\u00e7\u00e3o de pacientes do mundo real e incorpora dados de aproximadamente 1.000 variantes gen\u00f4micas candidatas. As amostras dos pacientes foram fornecidas como sangue com EDTA ou como manchas de sangue seco em cart\u00f5es de filtro usando o CentoCard\u00ae da CENTOGENE \u2014 uma solu\u00e7\u00e3o
